Check that the vacuum line to the petcock is connected.

With the bike off and on the stand, turn the rear wheel slowly and check for tight spot/s in the chain.

A screw drive will work (long one works better). Not a good a stethoscope but works in a pinch. Just put the handle end on your ear and the other in on the engine and start searching for the loudest...

Could be dirty brake fluid and/or the caliper is not moving freely. Would suggest removing caliper and cleaning and replacing brake fluid. May not fix your problem but easy and cheap to do.
